    Stay Trippy: Cancer’s Rollercoaster

    Juicy J loves to stay trippy, and so does cancer. Cancer is really weird; what it does to a body in its entirety is rooted in what happens at the molecular level. People have had ideas to explain the behavior of this disease, some which have been dismissed as wacky, while other ideas have stood the test of time.

    Interestingly, the Warburg hypothesis, developed by Otto Warburg, has undergone a rollercoaster effect.

    Warburg observed something very unique about cancer cells in that they do not fully break down glucose (sugar) to generate the full potential of ATP molecules. In fact, cancer cells are extremely “inefficient” at breaking down sugar and produce lactic acid as a byproduct. Yeah, that thing your muscles produce when they are deprived of oxygen! What’s interesting is that cancer cells produce this even when oxygen is rich. Since a fully functioning mitochondria, or “powerhouse of the cell” is imperative to full breakdown of glucose, Warburg postulated that the lack of full glucose breakdown, or the accumulation of lactic acid, was a result of a mitochondria gone bad in cancer. That’s it! He solved the riddle! Mitochondrial dysfunction was the cause of cancer!

    Well, unfortunately that was not the case (at that time), and what he observed was not a cause of cancer, but more a passenger effect of the disease. What science later discovered was that a main driver of cancer was the activation of genes that promote uncontrollable growth, or here’s a riddle, the deactivation of negative regulator of growth (a double negative). So, for years after Warburg’s theory, the scientific community, alongside advances in genetics became obsessed with finding that magical gene/protein that we could target chemically and save the world once and for all.

    Turns out it’s not that simple either. What’s a hit for one cancer type may be irrelevant to another, and even these genes/proteins that we identify as a hit, need to be working alongside another wacky component of the cell to actually get a cancer rolling. A perfect storm…for the Warburg hypothesis. Interestingly, the scientific community didn’t really give a shit about Warburg’s theory until about 15 years ago when all of a sudden, the role of the mitochondria in cancer began to resurface. Now there are drug companies aiming to target that magical mitochondrial protein in cancer. I even attended a conference in Snowbird, Utah recently (go there if you love to ski btw), that was entitled “tumor metabolism.” It attracted many of the world’s rock star scientists (I was just a peasant in the crowd), but we all came together as a scientific community, and the recurring theme was…targeting the mitochondria in cancer. How happy Otto Warburg would be.

    Peroxisomes

    In 1954, James Badhouin, a Ph.D. student at the Karolinska Institute in Sweden, discovered a tiny spherical compartment of the cell that displayed unique biochemical properties. Badhouin appropriately termed this the “microbody” but despite the fact that it housed enzymes, it was perceived in the literature as merely “junk” within the cell. This piqued the interest of future Nobel Laureate Christian de Duve, who identified that a key detoxification enzyme, catalase, resides within microbodies. The job of catalase is to decompose the toxic hydrogen peroxide to non-toxic water and oxygen; the term “peroxisome” was appropriately used to replace microbody.

    Fast forward 20 years, and the research community still believed peroxisomes were junk components in the cell. However, a series of biochemical and pathological experiments in the 1980s found that mutations in DNA encoding for peroxisomes actually resulted in severe childhood disorders known as Zellweger spectrum disorders (ZSD). Thus, peroxisomes were not junk but served an important biological function. Specifically, ZSDs were characterized by an accumulation of distinct types of fats called very-long chain fatty acids (VLCFAs). It was later discovered that peroxisomes were responsible for converting VLCFAs into shorter fats which could be used for key cellular signaling and metabolic processes.

    As a cancer researcher, when I explain what peroxisomes are to people, they often still refer to them as junk, or ask me what does that even have to do with cancer? I thrive under such scrutiny. Cancer is a disease characterized by disordered metabolic wiring and cellular signaling, and the peroxisome is a bona fide metabolic and signaling hub within the cell.

    My research has recently found that peroxisomes were elevated in chemotherapy resistant cancer cells cultured in a petri dish, and even in patients who don’t respond to chemotherapy. This is an association however, but the best way to figure out if something is contributing to a process is to mess with it.

    For instance, let’s say there was this disc on two wheels of a car and you didn’t know what it did. You could remove it, and discover that the car would no longer be able to stop. Or maybe you could add discs to the other wheels, or better discs, and find that the car stops more efficiently. Well that’s the same principle behind what I did to peroxisomes in cancer cells. I literally removed them and found that the cells lost their resistance to chemotherapy. Even in cells that do respond to chemotherapy, when I remove peroxisomes and add chemo, the cells die, but way more than they do with chemo alone.

    Currently there are no drugs that target peroxisomes in cancer, but hopefully my research will lead to this one day. It is also important to note that there are many efforts being made to use drugs to promote peroxisome function in childhood disorders. Such investigations are equally important. What we do know about drug screens though is that if we have a panel of say 1000 drug candidates, some will improve function, and some will do the opposite of what we intend (and vice versa).

    So, perhaps in drug screens that are aimed to improve peroxisome function in young adults, we may stumble across some that do the opposite. Conversely, if we screened for drugs that break peroxisome function in cancer maybe we would come across some that improve it. One field’s junk could be another field’s treasure. So, what have I learned from all this so far? Go against the grain, listen to those who doubt you, and if it aint’ broken….break it.

    NBA Playoff’s 2018 Has Begun

    As of this weekend the NBA playoffs just started. Sucks that its during one of the busiest weeks of my life but that’s neither here nor there. The last two years have been quite boring as the finals have been the same three teams for the last three years. Cleveland Cavs and the Golden State Warriors.

    Due to a ton of injuries and trades the road to the finals is uncertain for any team. Boston  is already short-handed without superstars Kyrie and Eric Gordon (out) have beaten the Bucks 2-0 in their series. Toronto has Washington right where they want them in there series up 2-0.

    The second match up between the Pacers and the Cavs could prove interesting. Seeing how the Cavs were unprepared for their game one loss. The should be ready to play Wednesday night. Houston “barely” made it past the Timberwolves defense and the series may go to 6 or 7 games after the nail bitter from Sunday night.

    Giving space for new life to run its course

    My heart is full

    My veins are full

    With blood that flows through them

    That blood that keeps me so warm

    The leaves and their trees can be accountable for

    As they ooze out oxygen, I breathe

    Without them there would be no me

    Not only is my life like a leaf

This is huge! As a Dominican environmentalists @angiem_ao and someone who grew up around this area. Seen this 😭brought tears of joy! Also swipe left, 🤗 I’m sure this is a collective joy and a sign of hope for those who live around this river . 

The Ozama River is heavily polluted. It is constantly affected by the slums on its shores and the factories that dump their waste into it. It is one of the main causes of pollution on the coastline of Santo Domingo. One of the many reason I’ve decided to be an #enviromentalscientist 

It’s also one of the areas most affected right now with COVID-19 😔

So grateful @boyanslat and @theoceancleanup 🌻 for the work you do in keeping our 🌎 oceans clean and making sure that areas of the most marginalized communities living near rivers or the beach have less pollution = cleaner, educate locals which creates more sustainable conscious communities for the future generations. ✊🏾#betheexampleyouwanttosee 🌻

There are literally people comfortable in their home with everything paid for them. Shaming accounts who are voicing THE TRUTH and commenting “they deserve what they get” 🤬

White privilege also is rooted from capitalism and instead of using your privilege for good. You are being violent and creating more #inequality while most of us are trying to break this mental insanity created by greedy colonizers which most of these descended people are highly benefiting from.

    Most of us are still processing and healing racial Most of us are still processing and healing racial trauma and the shock of seen white supremacy and racism at work on January 6,2020 in the US. 

However, just because is uncomfortable and seen the truth plus the reality that POC are dealing on a daily basis. Now, at night during lockdown “curfew” in Montreal, Quebec 🤦🏽‍♀️ 
we must speak about important matters. We don’t want to deal with bigger problems down the line. 

Just like MLK 🖤✊🏾once said: “Injustice anywhere is a threat to justice everywhere. We are caught in an inescapable network of mutuality, tied in a single garment of destiny. Whatever affects one directly, affects all indirectly."

Their actions and decisions are really detrimental and silent violence that only people who are in the front line, essential workers, teachers, children, BIPOC communities, Women of color, Parents, Trans, LGBTQ+, homeless people and elderly are suffering the most. I would not be surprised these leaders of this city are very supportive of these groups. Stay conscious and pay attention. 🌻

We must not stay silent #canadians don’t think that what happen in the US cannot happen here. And they’re counting on your silence and complicit so they can continue their toxic behaviours. It has to end and it ends with all of us, not just some of us.
